At the 2025 National Palace Museum Southern Branch Marathon, the runner tackled their first full marathon, moving from one aid station to the next through late fatigue. The start and finish are next to the Celebration Garden on the north side of the Southern Branch of the National Palace Museum in Taibao City,
Chiayi County, and based on the previous edition, the course was laid out as a large clockwise loop around the museum.
It passes access roads to the museum such as Gugong Beilu, Gaotie Dadao (高鐵大道, High Speed Rail Boulevard), and Gugong Dadao,
continues onto the cycling/walking path atop the Puzi River levee,
and also passes through the chinaberry tree tunnel that is a signature feature of the race.
It's known for running through Chiayi's southern plains, where farmland and river levees come together.

regular daily free shuttle also runs Tue-Sun 9:00-17:30 every 20 min from HSR Chiayi Station Exit 3 to the museum's south gate bus stop.

Entry fee fees are NT$1,300 for full marathon, NT$1,150 for half marathon, NT$1,000 for 10.7km,
NT$800 for 7km, and NT$700 for 3km (run/walk), with an additional NT$100 chip deposit for each.
Each category is capped at 1,000 participants,
and the time limit limits are 7 hours for full marathon, 4 hours for half marathon, 3 hours for 10.7km, and 2 hours each for 7km and 3km.
Bib and the participant kit, including a commemorative jacket, are mailed to runners' homes 1–2 weeks before the race.
On race day, a free shuttle bus runs from Chiayi HSR Station to the event venue,
and presenting bib allows runners to exchange it for free admission to the National Palace Museum Southern Branch.
